Immunoprecipitation consists in protein purification from a cell extract to isolate a specific protein, through the interacyion of an antibody with the protein of interest.

It can be executed by indirect method, or direct method.

Indirect: after incubation, the antibody/antigen complex is pulled out of the sample by using protein A or protein G-coupled agarose or magnetic beads. This allows the isolation of the protein from the sample, which then can be analyzed by western blot, mass spectrometry, ELISA, or other techniques.

Direct: Agarose Antibody Conjugates, or specific affinity beads can be used to eliminate reagents such as Protein A and Protein G and allow a ONE-STEP incubation for IP.

Protein agarose for specific purification of immunoglobulin is available as either Protein A, Protein G, Protein G-PLUS or Protein L conjugated to agarose and pre-treated to reduce non-specific immunoglobulin binding.
